Methodological study on assay of netilmicin concentration in body fluid

Abstract
A microbiological method has been established for assay of domestic netilmicin concentration in body fluid. By using this method, the sensitivity, lowest concentration and reabsorption rate reached 0.0375~0.0751��g, 0.25~0.5��g/ml and 88.8% ��3.63%~98%��14.40% respectively. The relative standard difference (RSD) of repeated test was 2.22%~5.34%. One hundred blood samples from 10 healthy volunteers have been assayed by this method and HPLC. The blood concentration determined by former method was much higher than by the later one (P < 0.01). The concentration lowered by HPLC was due to the need of complicated pretreatment of samples and specific chromatographic condition. The reabsorption rate of HLPC was only 71.98%~76.96%. However, the correlation between the results of the two methods was satisfactory. Results suggested that microbiological method with the advantages of convenience, simplicity, high accuracy and reliability can be used to assay the netilmicin concentration in body fluid.
