核心蛋白聚糖对HepG2细胞表皮生长因子受体转导蛋白及Ras/Raf信号通路的影响

Abstract
目的将核心蛋白聚糖(DCN)真核表达载体转染到HepG2中并检测其表皮生长因子受体(EGFR)表达。方法经大肠杆菌JM109介导DCN真核表达载体转染HepG2细胞,建立稳定转染的细胞株,采用Western印迹法和免疫组化检测其EGFR表达。结果 pFRT/laczeo-DCNHepG2细胞中EGFR较pFRT/laczeo HepG2明显高表达;pFRT/laczeo-DCN HepG2细胞中EGFR颗粒明显减少、变小。结论 DCN转染的HepG2细胞TGFR减少,能够抑制MAPK/Ras/Raf信号通路,从而阻止、抑制HepG2的生长。
