应用抑制消减杂交技术筛选大肠癌相关基因

Abstract
大肠癌是我国常见的消化道恶性肿瘤,其死亡率也呈增长趋势.研究显示,大肠癌发生过程是一个多步骤、多因素、涉及到多种基因及产物的相互作用.但至今尚未发现与大肠癌发生发展有关的特异性基因.本课题利用抑制消减杂交(suppression subtractive hybridization,SSH)构建大肠癌与癌旁组织的消减cDNA文库,系统分离差异表达基因,并分析这些基因的功能,以期筛选到新大肠癌相关基因,为进一步明确大肠癌发生发展的分子基础及临床大肠癌的诊治提供研究基础.
