Inactivation effect of photochemical treatment by methylene blue derivative on human osteosarcoma cell line

Abstract
Objective To study the inactivation effect of different concentrations of methylene derivative(M007) on human carcinoma(MG-63) cells,and to provide essential information for tumor cell inactivation in intraoperative blood salvage.Methods Study groups were divided into Group C,Group P,GroupM007 and Group M007-PCT.106 cells/ml MG-63 cells and different concentrations of M007 were added into culture dishes.After shaking and irradiation by red visible light,the death of suspended cells was detected by the trypan blue and examined under microscope.Results In Group M007-PCT,MG-63 cell deaths increased with increasing concentrations [(2~4)μmol/L]of M007(P0.05).When the dose of M007 was(4~32)μmol/L,the death rate of human Osteosarcoma cells reached a high level(93%) and didn't increase with further increase of M007(P0.05).The cells cultured with M007 but without irradiation showed a little increase in death rates with increasing concentrations of of M007,but the death rates were much lower than those of the M007-PCT group(P0.05).Conclusion The results show that M007 can effectually inactivate human Osteosarcoma cells,which makes this new photosensitizer a promising agent with potential application in blood salvage.
