Study on extraction of active polysaccharides from Ganoderma lucidum

Objective To explore an optimum extraction procedure(water extraction-alcohol precipitation and ultrasonic extraction method) of active polysaccharides from Ganoderma lucidum.Methods By using colorimetric methyl thiazolyl tetrazolium (MTT) assay of splenic cells of mice as evaluation indicator,orthogonal test was conducted to evaluate ultrasonic extraction time,water extraction temperature,and water extraction time to explore the optimal extraction procedure.Results The main influencing factors of colorimetric methyl thiazolyl tetrazolium (MTT) assay of splenic cells of mice were ultrasonic extraction time,water extraction temperature,and water extraction time in turn. The optimal conditions of orthogonal experiment:ultrasonic extraction for 20 minutes,water extraction at 80 ℃ for 2 hours. Ultrasonic extraction time had significant effect on extraction of active polysaccharides from Ganoderma lucidum.Conclusion The technology provided references for extraction of active polysaccharides from Ganoderma lucidum.
