利用紫外诱变绿色木霉对玉米秸秆进行酶解糖化的研究

Abstract
以绿色木霉菌株为出发菌株,通过紫外诱变,筛选出纤维素酶产量高且具有遗传稳定性的菌株Z;在经稀硫酸预处理后的玉米秸秆中加入菌株Z进行酶解糖化试验。结果表明:在紫外线强度15W,照射距离30cm,照射时间60s的条件下,绿色木霉的致死率为94.1%,在此条件下选取菌株Z,具有良好的遗传稳定性,酶活为37U.mL-1,比诱变前酶活提高12%。利用该菌株,在酶解糖化时间60h、加酶量45U.g-1底物、底物浓度105g.L-1、温度37℃、初始pH值4.8时,还原糖得率为30.02%,说明绿色木霉经过诱变产纤维素酶具有较强的活性。
