Abstract
在前期研究基础上,采用复乳化法制备载牛血红蛋白纳米微球型血液代用品。免去传统的匀浆强力分散措施,通过表面活性剂的加入及溶剂筛选,在初乳和复乳步骤均采用较温和的超声波分散,获得97%以上的高包封率、平均粒径约286 nm微球。油相中表面活性剂亲水-亲油平衡值对包封率有显著影响,且与外水相表面活性剂匹配才能获得高的包封率。混合表面活性剂Poloxamer188/Span80比单一表面活性剂Span80能更稳定乳液,提高包封率。表面活性剂浓度高,有利于获得高的包封率和窄的粒径分布。但超过一定量时,对包封率影响不明显,而粒径及粒径分散指数增大。溶剂脱除速率适中有利于获得较高的包封率、较小的粒径和粒径分散指数。
