Abstract
目的 探讨磁性纳米Fe3O4颗粒(Fe3O4,-MNPs)对Mdr-1基因siRNA表达载体转染白血病耐药细胞株K562/A02细胞转染效率的影响.方法 构建靶向Mdr-1基因的PGCsilencer-U6-neo-GFPsiRNA表达载体(PGY1-1),分别与10、15、20μg/ml Fe3O4-MNP置于37℃、5%CO2条件下共孵育,荧光显微镜下计数48 h细胞的转染效率.结果 PGYl-1对K562/A02细胞转染率未见明显变化.结论 PGY1-1对K562/A02细胞的转染效率不能通过共孵育的方法得到提高.
