Abstract
Objective To establish an HPLC method for simultaneous determination of the contents of chlorogenic acid and caffeic acid in Solanum lyratum Thunb.Methods The operation was carried out on Diamonsil-C18 column(250 mm×4.6 mm,5 μm)with the mobile phase consisting of a mixture of methanol-0.05% phosphoric acid(V∶ V=25∶ 75).The flow rate was 1.0 mL · min-1,UV detection wavelength was set at 327 nm and the column temperature was 45 ℃.Results The calibration curve of chlorogenic acid was linear in the range of 0.5-32.0 mg · L-1(r=0.999 9).The mean recovery of chlorogenic acid was 99.9%,RSD was 1.52%.The calibration curve of caffeic acid was linear in the range of 10.0-640.0 μg · L-1(r=0.999 9).The mean recovery of caffeic acid was 99.9%and RSD was 1.35%.Conclusions The HPLC method is found to be simple,accurate,sensitive and reproducible.It can be used for the determination of the contents of chlorogenic acid and caffeic acid in Solanum lyratum Thunb.
