Abstract
目的 优化双向凝胶电泳的实验步骤,总结出一套适用于强直性脊柱炎的外周血CD4+ T细胞的双向凝胶电泳技术.方法 对双向电泳实验中的关键环节:样本处理,上样方法,聚焦条件等进行调整与优化,硝酸银染色后进行凝胶图像比较.结果 采用pH4-7胶条,用免疫磁珠分离外周血CD4+ T 淋巴细胞,Cleaning-up试剂盒沉淀蛋白,所得双向电泳图谱清晰,分辨率高,建立了稳定的2-D技术.结论 本文首次报道能得到约800个点的强直性脊柱炎的外周血CD4+T淋巴细胞的蛋白电泳图谱,为强直性脊柱炎的外周血CD4+T细胞蛋白组研究奠定了基础.
