VEGF121转染对诱导后血管内皮细胞增殖的影响

Abstract
目的 探讨VEGF基因转染对VEGF诱导后血管内皮细胞生物学功能的影响,为组织工程血管构建提供血管内皮细胞的可能性.方法 应用VEGF体外诱导兔骨髓间充质干细胞定向诱导分化成血管内皮细胞后,再通过脂质体介导的方法将pCDI/VEGF121转染分化后的血管内皮细胞,通过MTT及流式细胞仪检测转染前后细胞的增殖与凋亡能力.结果 诱导培养2周后兔骨髓基质细胞转化为血管内皮细胞,VEGF基因转染血管内皮细胞后表达VEGF蛋白,转染后细胞增殖能力增加,凋亡能力下降.结论 转染VEGF是促进VEGF诱导血管内皮细胞增殖的有效途径,可望为组织工程血管构建提供血管内皮细胞衬层,为人工血管内皮化开辟一种新的途径.
