Effects of mitomycin on the morphology of human gastric cancer cells and GSTΠ expression

Objective: To study the effects of mitomycin on the morphology of human gastric tumor cells and GSTΠ expression. Method: Mitomycin was added to human gastric tumor cells cultures, and kept for 5h and 10h respectively. The gastric cancer cells were observed under optical microscope and transmission electron microscope and stained with immunohistochemical method. Result: For the 5-hour-treatment group, the morphology of gastric cancer cell did not changed significantly, but GSTΠ expression was reduced in cytoplasm and increased in nuclear; for the 10-hour-treatment group, the organelles were apparently destroyed, the cell membranes were burst, the expression of GSTΠ was little in cytoplasm and nuclear. Conclusion: Mitomycin has activity of killing gastric cancer cells, and it maybe relate to inhibition of the expression of GSTΠ gene.
