积雪草酸对肝癌细胞增殖作用的影响

Abstract
目的:研究积雪草酸(asiatic acid,AA)对人肝癌细胞株(HepG2)的抑制作用,并探讨其与线粒体之间的关系。方法:采用MTT法检测AA对HepG2细胞增殖的影响,倒置显微镜观察细胞形态的变化;荧光探针MitoTracker检测线粒体数目;荧光探针JC-1检测线粒体膜电位;荧光探针DCFH-DA检测胞内活性氧(reactive oxygen species,ROS)含量;ATP试剂盒检测胞内ATP水平;RT-PCR和Western Blot法分析电压依赖性阴离子通道(voltage-dependentanion channel,VDAC)表达量的变化。结果:AA能够剂量依赖性抑制肿瘤细胞株的增殖,30μmol/L AA作用细胞24h后细胞突起消失、胞体皱缩、变圆。AA能够使HepG2细胞线粒体膜电位下降,胞内ROS含量增加,降低胞内ATP水平,50μmol/L AA能够增加VDAC蛋白的表达量。结论:AA抑制肿瘤细胞增殖的作用与线粒体有关,线粒体VDAC参与人肝癌细胞凋亡的调控过程。
