高糖对HSkMCs细胞株AT1R、mfn2mRNA表达的影响

Abstract
目的探讨骨骼肌局部肾素血管紧张素系统(RAS)在高糖状态的变化及对线粒体融合蛋白2(mfn2)基因表达的影响。方法体外培养人骨骼肌细胞(HSkMC),分为不同浓度葡萄糖组进行培养:5.55mmol/L组,11.1mmol/L组,22.2mmol/L组,分别培养48h,应用RT-PCR检测各组的血管紧张素Ⅱ-1型受体(AT1R)、mfn2基因表达。结果以5.55mmol/L组作为基础对照组,11.1mmol/L组、22.2mmol/L组AT1R mRNA表达均较对照组增加(P<0.05),而mfn2基因表达则下降(P<0.05)。结论高糖能诱导骨骼肌细胞AT1RmRNA表达上调,mfn2mRNA表达下调,且呈剂量依耐性。
