Abstract
Objective To explore the possible reverse effect and the mechanism of on a (Verapami Vera) human tongue cancer drug-resistant cell line Tca8113/BLM in vitro. Methods The reverse effect of Vera on Tca8113/BLM was determined by MTT. The effects of Vera on cells ADM congregation,and the expression of P-gp and MRP were detected by flow cytometry (FCM),The cell apoptosis were observed by laser confocus microscope(LCM). Results The value of IC50 of Tca8113 and Tca8113/BLM cells without Vera were 16.1 and 86.73,respectively. While after treated with Vera (10μmol/L),The value of IC50 of Tca8113 and Tca8113/BLM cells were 8.1 and 8.06,respectively. the potent folds of BLM inTca8113/BLM cells were 12,and the concentration of ADM in Tca8113/BLM cells increased obviously(P 0.01).and the apoptosis of Tca8113/BLM were obvious,And the expression level of P-gp in Tca8113 andTca8113/BLM cells treated by Vera decreased significantly. Conclusion Vera can increase the intracellular concentration of ADM in Tca8113/BLM cells and reverse the drug-resistance of Tca8113/BLM cells to BLM. The reverse effect is possibly associated with the combination of Vera and P-gp.
