Abstract
Objective To investigate the effects of PEP-1 mediated heme oxygenase-1(PEP-1-HO-1)fusion protein on TNF-α mRNA expression in liver of rats with ischemia-reperfusion injury. Methods SD rats were respectively injected HO-1(HO-1 group) or PEP-1-HO-1(PEP-1-HO-1 group) with dose of 300 μg in tail vein, then the liver ischemia-reperfusion injury model were prepared in these rats, and control group was designed. The TNF-mRNA expression levels in liver were detected with RT-PCR at 1,6,12,24 h,respectively. Results The TNF-α mRNA expression in PEP-1-HO-1 group were significantly lower than those of HO-1 group and IRI group(P0.05),and the expression was decreased following the injury prolong(P0.05).Conclusion PEP-1-HO-1 fusion protein could decrease the expression of TNF-α mRNA,so as to lighten liver ischemia-reperfusion injury.
