Abstract
目的研究Gravin在血管内皮生长因子(VEGF)诱导人脐静脉内皮细胞(HUVECs)迁移和体外"小管样结构"(TLS)形成中的作用。方法用VEGF诱导HUVEC,通过Western blot和细胞免疫荧光分析Gravin在细胞内分布的变化;用G(o|¨)6983【蛋白激酶C(PKC)抑制剂】和VEGF共处理内皮细胞,观察对Gravin分布的影响,观察其对VEGF诱导HUVECs迁移和TLS形成的影响。结果VEGF呈时间和剂量依赖性诱导Gravin磷酸化,并向核周和细胞边缘重分布;以上效应能被0.5μmol/LG(o|¨)6983抑制;G(o|¨)6983同时抑制VEGF诱导HUVEC迁移和TLS形成。结论PKC-Gravin信号通路参与VEGF诱导的内皮细胞迁移和血管形成。
