Abstract
目的探讨神经生长因子对β-淀粉样蛋白诱导神经元内酸碱度及凋亡的影响。方法培养大鼠大脑皮质神经元细胞,应用Aβ25-35构建AD细胞模型,并加入神经生长因子干预,荧光探针法检测细胞内pH值,West-ern印迹分析法测NHE1及CaM表达。结果经Aβ25-35诱导的AD细胞模型NHE1表达抑制,细胞内pH降低,神经生长因子有效增强NHE1表达(P<0.01),提高细胞内pH(P<0.01),下调CaM表达,对抗Aβ25-35的毒性。结论细胞内酸化在神经元凋亡中起重要作用,神经生长因子可增强NHE1表达、提高细胞内pH值,达到抗细胞凋亡的目的 。
