Abstract
目的 探讨蒺藜皂苷(GSTT)对H2O2诱导的PC12细胞氧化应激损伤的保护作用.方法 用H2O2刺激PC12细胞使其发生氧化应激损伤,MTT法检测PC12细胞活性,流式细胞仪检测PC12细胞凋亡率及线粒体膜电位变化,Hoechst33258染色观察PC12细胞核形态,琼脂糖凝胶电泳观察DNA ladder图谱.结果 与模型组比较,GSTT各剂量组可使PC12细胞存活率提高(P<0.001,P<0.01),凋亡率降低,线粒体膜电位回升,细胞核浓染致密数减少,无典型DNA ladder条带,且在一定范围呈剂量依赖性.结论 蒺藜皂苷对H2O2诱导的PC12细胞氧化应激损伤具有保护作用,其作用机制可能与稳定线粒体膜电位有关.
