Abstract
Objetive Research whether the Hep-2 tumor vaccine with B7-1 can improve CIK cells' specific immunity. Mothods To co-cultured CIK cells and Hep-2 cells transfected with B7-1 and determined CIK cells' the growth rate, Phenotypes and Cytotoxicity.Results After co-cultured 21d, the growth rate of transfected group was significantly proliferated compared with that of control group(p<0.01),whereas no-transfected group had a similar growth rate compared with control group .The percentage of CD3+CD56+ cells and CD3+CD8+ cells of transfected group was significantly higher than that of control group (p<0.01), whereas the other two groups had similar growth rate, In Cytotoxicity assay, when the effector cells and target cells were co-cultured at ratio of 40:1, different CIK cells showed high cytolytic activity in different time . When Hep-2 cells were used as target cells, transfected group cells showed significantly higher cytolytic activity compared with that of control group(p<0.01,two-tailed t-test),whereas the other group cells had asimilar cytolytic activity. When K562 cells were used as target cells, each group all showed relatively similar cytolytic activity (p>0.05). Conclusions Co-culturing Hep-2 tumor vaccine with B7-1 and CIK cells may increase CIK cells' cytotoxicity to tumor cells.
