Abstract
目的:建立大鼠血浆中氯吡格雷羧酸代谢物SR26334检测的高效液相色谱方法,研究SR26334的药代动力学。方法:血浆经高氯酸沉淀,以ZORBAX SB-C18为色谱柱;流动相为乙腈-水-0.1%TFA(三氟乙酸),流速为1.0 ml/min;检测波长为230 nm(0 min~4.5 min)、280 nm(4.5 min~6 min)。结果:SR26334浓度在0.5 mg/L~50 mg/L范围内线性关系良好(r=0.9999);定量下限为0.5 mg/L;低、中、高三个浓度的相对回收率100.34%~101.78%;日内RSD(相对标准偏差)和日间RSD均小于10%。结论:本方法准确可靠、简便快速,适用于大鼠血浆SR26334浓度的测定。
