Abstract
【Objective】 Peach(Prunus persica Batsch) ceases growth and becomes dormant during winter to elude the detrimental effect of exposure to low temperatures.This state has been designated endodormancy.To better understand the genetic control of flower bud dormancy in peach and select the related gene,【Method】 We identified differentially expressed genes during dormancy progression in flower buds from medium chilling requirement peach variety 'Mantianhong' by using cDNA-AFLP.【Result】 Twelve primer combinations were used for selective amplification.Five hunderd and twenty-five TDFs(transcripts-derived fragments) were obtained for their differential expression during dormancy release.Among the 525 TDFs,14 TDFs were up-regulated and 18 TDFs were suppressed.Fourteen up-regulated TDFs were cloned and sequenced.Ten TDFs presented high similarity to known genes which encoded E3 ubiquitin-protein ligase CIP8,Histone H3.3,60S ribosomal protein L34,Prohibitin-2,and Galactinol-sucrose galactosyltransferase 2.【Conclusion】 The TDFs whose functions are involved in metabolism,stress response,cell structure and plant growth and signal transit according to their primary BLAST match could help us to understand the mechanism of bud dormancy in peach.
