Synthesis of ~(13)C_6-1,2:5,6-Di-O-isopropylidene-α-D-glucofuranose

With concentrated sulfuric acid as the catalytic agent,13C6-1,2:5,6-Di-O-isopropylidene-α-D-glucofuranose was synthesized with D-glucose-13C6 and acetone.13C labeled 1,2:5,6-Di-O-isopropylidene-α-D-glucofuranose was characterized by GC and MS.The reaction conditions were optimized to be as follows: the molar ratio of [D-glucose-13C6]∶[acetone] = 1∶73.5,the volume of concentrated sulfuric acid = 10 mL,the reaction temperature = 25 ℃,and the reaction time = 12 h.Under these conditions,the yield of 13C6-1,2:5,6-Di-O-isopropylidene-α-D-glucofuranose was 75.6%(based on D-glucose-13C6),with 99.2% 13C atom abundance and 98.7% chemical purity.
