Abstract
目的 研究阻断鞘氨醇激酶1 (sphingosine kinase 1,SphK1)通路对高糖诱导的大鼠系膜细胞(glomerular mesangial cells,GMC)中纤维连接蛋白(fibronectin,FN)表达的抑制作用.方法 采用Realtime PCR检测SphK mRNA水平;Western blot检测FN的蛋白表达.结果 SphK1抑制剂二甲基鞘氨醇(N,N-Dimethylsphingosine,DMS)显著降低高糖诱导的FN表达;转染SphK1 siRNA可有效消除高糖诱导的FN上调效应.结论 SphK1通路在高糖诱导的肾小球系膜细胞FN表达过程中发挥着重要的调节作用.
