甲基强的松龙抑制实验性自身免疫性脑脊髓炎IL-23/IL-17轴机制研究

Abstract
目的 探讨甲基强的松龙(methylprednisolone, MP)对大鼠实验性自身免疫性脑脊髓炎(experimental autoimmune encephalomyelitis, EAE)新发现的IL-23/IL-17炎性轴发病机制的影响.方法 建立Wistar大鼠EAE动物模型,给予糖皮质激素MP和生理盐水预处理建立MP组和EAE组,并设立正常对照组,RT-PCR法检测大鼠脑组织中白介素-12(interleukin-12,IL-12)和白介素-23(interleukin-23,IL-23)亚基IL-23P19、IL-12P40、IL-12P35及白介素-17(interleukin-17,IL-17)前体毒性T细胞相关抗原8(cytotoxic T lymphocyte-associated antigen 8, CTLA-8)的mRNA表达,用ELISA法检测IL-17水平.结果 MP组人鼠脑组织中IL-23P19、IL-12P40、IL-12P35及CTLA-8的mRNA的表达比EAE组低(P<0.05),IL-17水平也低于EAE组(P<0.05),差异均有统计学意义.结论 甲基强的松龙能够抑制IL-23/IL-17轴而起到抗炎治疗作用.
