In Vitro Anti-tumor Activity of Glycosides from Total Steroidal Glycosides of Radix Cynanchi Auriculati

Abstract
Objective To separate and purify the glycosides from the root of Cynanchum auriculatum Royle ex Wight and investigate the anti-tumor effect of it on the multiform tumor cells. Methods The anti-proliferation effects of glycosides on B-16、K-562、PC-3 and Hela tumor cells were studied by MTT assay,growth curve methods and cell morphological observation. Results Glycosides of Radix Cynanchi Auriculati had potent cytotoxic effect on all four tumor cells.The 50% inhibiting concentration(IC50) of which were(10.0±0.4),(23.1±1.3),(34.8±2.4),(41.6± 6.4) μg·mL-1 respectively,showing evident dose-effect relationship.It was indicated that the anti-tumor effect of glycosides on PC-3 tumor cells enhanced as the concentration increased at a certain time and as the time increased in a certain range of dosage by growth curve methods.The glycosides of Radix Cynanchi Auriculati significantly inhibited the growth of tumor cells,reduced density of tumor cells in culture media,where some cells crimpled,some suspended,and some decomposed into fragments. Conclusion The glycosides of Radix Cynanchi Auriculati could inhibit the growth of many kinds of tumors in vitro and was selective to certain kinds of tumors.
