高山蓍嫩茎无性系的建立

Abstract
【目的】建立高山蓍嫩茎再生繁殖体系,满足高山蓍人工栽培对种苗的需求,保护其野生种质资源。【方法】以嫩茎为外植体,分别探讨不同培养基和激素组合对愈伤组织的诱导、分化、不定芽生根、试管苗继代增殖培养的影响。【结果】在添加不同激素组合的MS、1/2MS、8114和B5培养基中,高山蓍嫩茎愈伤组织诱导培养的理想培养基为1/2MS+6-BA0.8mg/L+NAA0.5mg/L+2,4-D1.5mg/L和1/2MS+6-BA0.8mg/L+NAA0.5mg/L+2,4-D2.0mg/L;愈伤组织分化培养的理想培养基为1/3MS和1/3MS+IAA0.2mg/L;在添加NAA0.6mg/L或IAA0.6mg/L的1/2MS、1/4MS、N6、B5和LS培养基中,1/4MS+IAA0.3mg/L是不定芽生根培养和试管苗继代增殖的理想培养基,生根率为92%,平均每代繁殖系数为2.7;试管苗易移栽定植,成活率可达98.6%。【结论】建立了高山蓍嫩茎繁殖无性系,定植的试管苗生长旺盛,翌年开花结果,并保持了高山蓍的所有植物学性状。
