Abstract
目的观察眼镜蛇毒因子(CVF)对慢性坐骨神经损伤(CCI)后大鼠脊髓背角神经元的保护效应。方法 36只SD大鼠随机均分成三组,CCI+生理盐水(CS组)、CCI+CVF 50μg/kg(CC组)和尾静脉注射生理盐水(SS组)。于术前和术后1、3、5、7 d测定大鼠的热痛阈值和机械痛阈值,并于术后7 d处死大鼠取脊髓组织,在电镜下观察脊髓背角神经元的内部结构变化,并测定脊髓匀浆中超氧化物歧化酶(SOD)和丙二醛(MDA)吸光度。结果与SS组相比,CS组大鼠热痛阈值和机械痛阈值降低,脊髓匀浆中SOD活力明显降低,而MDA显著升高(P<0.05);与CS组相比,CC组大鼠热痛阈值和机械痛阈值升高,脊髓匀浆中SOD活力明显升高,而MDA却迅速下降(P<0.01)。给予CVF后,CCI大鼠脊髓背角神经元线粒体的肿胀程度减轻。结论 CVF能减轻CCI后脊髓背角神经元的损伤程度,可能与其减少自由基和脂质过氧化的损伤有关。
