Dna Vaccine Restores Hearing Loss Induced By Beta-Tubulin In Guinea Pig

RATIONALE: DNA vaccine has been shown to be effetive in the treatment of immune disorders. We want to restore hearing loss induced by beta-tubulin in guinea pig using DNA vacine. METHODS: Female Hartley guinea pigs(350-450g) were used in this study. All animals were immunized by 200 ug tubulin mixed with the same volume of complete Freund's adjuvant. The tubuin was subcautaneously injected in the back of the guinea pigs. The immunization was booosted once with incomplete Freund's adjuvant (IFA) 1 week later (total dose was 400 ug). Hearing tests was conducted before and after the immunization. After the second hearing test, 250 ug control and DNA vaccine was injected into the guinea pig right thigh intramuscularly. After 1 weeks, then we performed hearing test again. RESULTS: Before immunization, the threshold of guinea pig was 55dB SPL and after immunization the hearing threshold was 85dB SPL and vector control was 78dB SPL and guinea pig immunized with DNA vaccine was 55dB SPL. CONCLUSIONS: DNA vaccine could revert autoimmune inner ear disease induced by auto-antigen β-Tubulin and restored hearing loss in guinea pig.
