Effect of butylphthalide on onexpression of aquaporin-4in cortex of rats following cerebral ischemia

Objective To study the effect of butylphthalide on onexpression of aquaporin-4in cortex of rats following cerebral ischemia and its mechanismin protecting rats against cerebral ischemic injury.Methods One hundred and forty male SD rats were included inthis study.A global cerebral ischemia model of rats was established by ligating 4blood vessels.The 72rats used in establishing the model were randomly divided into cerebral ischemia group,butylphthalide treatment group and sham-operation group ( 24in each group ) .The rats in butylphthalide treatment group were further divied into 0.3mg / kg group,1.0mg / kg group and 3.0mg / kg group ( 8in each group ) .The Evans blue and water level and aquaporin-4expression level in brain tissue were measured by immunohistochemistry.Results The Evans blue and water levels were significantly higher whereas the aquaporin-4expression level in brain tissue was significantly lower in cerebral ischemia group than in sham-operation group group 1day and 1week after ischemia ( P0.05,P0.01 ) .The Evans blue and water levels were significantly lower whereas the aquaporin-4expression level in brain tissue was significantly higher in butylphthalide treatment group than in cerebral ischemia group 1day and 1week after ischemia ( P0.05,P0.01 ) .Conclusion The aquaporin-4is expressed in a time-dependent manner and butylphthalide can regulate the aguaprotein-4expression in rats following cerebral ischemia.
