Sialic Acid Analysis By Hpaec-Pad

The most labile monosaccharides are the family of sialic acids, which are usually chain-terminating substituents. These are therefore usually released first by either mild acid hydrolysis or enzyme digestion, and can be analyzed with great sensitivity by high pH anion-exchange chromatography with pulsed auperometric detection (HPAEC-PAD). The remaining oligosaccharide is analyzed as discussed in Chapter 103 to identify the position of linkage of the sialic acid.
