Abstract
An isolate from alkali soil of Tanggu,Tianjin with alkaline protease activity was identified by phenotypic classifica- tion and phylogenetic analysis.The results show that the strain HAP is a gram-postive bacillus with about(0.7-0.9)μm×(2-3)μm width×length.Many kinds of carbon sources and nitrogen sources could be utilized in the medium.Physiological and biochemical test showed that xylose,glucose,arabinose and mannose can be utilized by the strain as fermentable sugar for acid production.The strain could growth well in the substrates of casin and starch,and however,not in the substrate of tyrosine.It could utilize citrate,too.A se- quence alignment of 16s rDNA of the strain HAP was performed and the phylogenetic tree was constructed.The strain was identified as Bacillus alcalophilus.The protease activity of strain HAP is 1.22×10~4 U/mL.
