Determination of Amphotericin B in Human Serum by High-Performance Liquid Chromatography*: Bestimmung von Amphotericin B in Humanserum durch HPLC (high-performance liquid chromatography)

Summary: Amphotericin B in human serum is assayed after acetonitrile extraction by high‐performance liquid chromatography using methanol‐50 mM sodium succinate buffer pH 6‐dimethylsulfoxide (75:20:5 V/V) as the mobile phase; within a range of 0.1 to 10 mg/I the peaks heights are linearly related to the amphotericin B concentration. The other most employed antifungal drugs namely 5‐fluorocytosine and ketoconazole do not interfere in the assay. Zusammenfassung: Amphotericin B wurde in Humanserum nach Acetonitril‐Extraktion mit Hilfe der high‐performance liquid chromatography bestimmt. Als mobile Phase wurde ein Gemisch aus Methanol, 50 mM Natriumsuccinatpuffer pH 6 und Dimethylsulfoxid im Verhältnis 75:20:5 (V/V) benutzt. Innerhalb eines Bereiches von 0,1 bis 10,0 mg/I steht die Höhe der Peaks in einer linearen Beziehung zur Amphotericin B‐Konzentration. Die beiden anderen am meisten angewendeten Antimykotika 5‐Fluorocytosin und Ketokonazol interferieren nicht mit dem Testsystem.
