Construction,expression,and identification of a recombinant immunotoxin hIL-2-Luffin P1

Objective To develop and identify a new recombinant immunotoxin hIL-2-Luffin P1 specifically killing tumor cells of cutaneous T cell lymphoma(CTCL).Methods A recombinant hIL-2-LuffinP1 DNA fragment was subcloned into an expression plasmid pET32a(+).The recombinant vector was identified by restriction enzyme digestion and DNA sequencing.After the recombinant vector was transformed into bacteria BL21(DE3),the expression of fusion protein hIL-2-Luffin P1 was induced by IPTG and identified by Western blotting using anti-His Ab and hIL-2 Ab.Results The new recombinant vector pET32a(+)-hIL-2-Luffin P1 was successfully constructed.The fusion protein of immunotoxin hIL-2-Luffin P1 was identified by Western blotting.Conclusion The success of constructing immunotoxin hIL-2-Luffin P1 will give a good experimental basis for further investigating its killing effect specifically against CTCL tumor cells.
