Inhibition effect of tetrandrine on division of fibroblast from keloid

Abstract
Objective:To probe the effect of tetrandrine on mitosis of scar fibroblast.Methods:Fibroblast was cultured with various concentrations of tetrandrine,then the cultured fibroblast samples were gathered on different days of time gradient.Mitosis index was determined by light microscopy.Cell cycle and its distributions were analyzed by flow cytometer.Results:Mitosis index of tetrandrine group was reduced obviously compared with blank group at all time points and the peak time was shortened in a dose-and time-dependent manner.In the flow cytometry analysis,it was found that tetrandrine could promote fibroblast to enter S and G2-M phase from G0-G1 phase and extend the time of S G2-M phase distinctly.Conclusion:Tetrandrine can block the division of fibroblast at G2-M phase in a dose-and time-dependent manner.
