Abstract
目的:研究锰超氧化物岐化酶模拟化合物(mimics of manganese superoxide dismutase,MnSODm)对人白血病K562/ADM耐药细胞增殖抑制及诱导凋亡作用.方法:以人白血病高表达耐药基因(mdr1)的K562/ADM多药耐药细胞为靶细胞,应用四氮唑蓝比色法(MTT法)测定细胞增殖活性;光学显微镜、荧光显微镜和透射电镜观察细胞形态学变化;Annexin v/PI双标记法检测K562/ADM细胞凋亡.结果:(1-50),mg/L MnSODm可抑制K562/ADM细胞增殖(P<0.01),并呈时间和浓度依赖性.MnSODm诱导48h后,K562/ADM细胞出现典型的凋亡形态学改变;Annexin V/PI双染显示凋亡细胞显著增高.结论:MnSODm体外明显抑制人白血病K562/ADM耐药细胞的增殖,并诱导K562/ADM细胞凋亡.
