Abstract
Die Synthese und neuropsychopharmakologische Prufung von sechs dopaminanalogen Derivaten des 4-Phenylpiperidins wird beschrieben. Der Einflus von Methylgruppen in α- und β-Stellung zum Dopaminstickstoff sowie der Einflus der Methylierung im Catecholteil des Molekuls auf das neuroleptische Wirkungsprofil wird diskutiert. Die sedierende Wirkung der Substanzen beginnt bei etwa 1.56 mg/kg. Die toxische Dosis ist 100 mg/kg. Die sterisch fixierten Cyclopropananaloga werden aus den entsprechenden trans-Enaminen und Diazomethan durch Katalyse mit Palladium(II)-acetat dargestellt. Sie sind uberraschenderweise wirkungslos.
Neuropsychotropic Activity of Dopamine Analogous Phenylpropyl- and Phenylcyclopropylpiperidines
Six new dopamine analogous derivatives of 4-phenylpiperidine were synthesized and examined for neuropsychopharmacological activity. The influence on the neuroleptic profile of methyl groups in α-or β-position relative to the dopamine nitrogen and in the catechol moiety is discussed. The central depressive activity of the substances ranges from 1.56 to 100 mg/kg. Some conformationally rigid cyclopropyl analogs were prepared by reacting the corresponding trans-enamines with diazomethane in the presence of palladium(II)acetate as a catalyst. The lack of activity in this series was unexpected.
