大孔吸附树脂分离纯化白芍芍药苷的工艺研究

Abstract
目的:筛选分离白芍芍药苷的最佳树脂,并对影响分离的各种因素进行系统研究,使分离工艺达到最优化.方法:采用静态与动态的吸附-解吸2种方法,以高效液相色谱(HPLC)法测定芍药苷的含量为评价指标,进行工艺筛选.结果:D101型大孔树脂分离效果最好,其最佳工艺为饱和动态吸附-洗脱量94.5mg/g(干树脂)、上样液芍药苷质量与树脂质量比1:10.5、上样液体积与树脂质量比(ml:g)10:1,上样液pH值6~7、以2~3ml/min的速率吸附,用3BV(树脂床体积)的水、以3~4ml/min的流速冲洗杂质,用3BV 40%乙醇、以2~3ml/min的流速洗脱.经D101处理后的芍药苷纯度和回收率均达90%.结论:该方法简单可行,分离效果好,能满足大生产的要求.
