Development and application of new method for semen cryopreservation in Tianshan wapiti

Abstract
Semen electroejaculated from 16 Tanshan wapiti stags and the different factors influencing their semen cryopreservation were explored.The factors include cryodiluent,freezing method and final concentration of glycerol.Based on the results,a new semen cryopreservation method has been developed,then motility,acrosome integrity and fertility of these frozen semen were evaluated.The results showed the best frequency of electroejaculation was 2 times each week.During the course of electroejaculation,voltage had risen year by year from 2003 to 2005,it has been proved that the patience of stags has existent.The seminal fluid quality appeared the best in October.During the experiment of cryopreservation,Tris buffer of 6% glycerol had the optimal effect,the motility of the thawed semen reached up to(48.14±13.15)%,acrosome integrity reached up to(56.34±14.84)%.126 Tanshan wapiti hinds which had been induced estrus synchronization and 107 Tanshan wapiti hinds whiched had normal estrus had been applied with these straws for artificial insemination,and the birthrate of first-cycle reached up to 50.80% and 78.50%,respectively.
