Abstract
目的制备具有软骨诱导活性的新型基因活性基质材料,以促进种子细胞定向分化。方法用固相法合成KLD-12多肽并通过高压液相色谱及质谱仪纯化并鉴定;取0.5%(w/v)多肽在生理溶液条件自组装成多肽凝胶状物质,将TGF-β3质粒与KLD-12多肽间非共价键结合负载质粒并检测其DNA释放能力。结果KLD-12多肽序列合成成功,其相对分子质量为1466.9 Da;携TGF-β3基因的自组装肽凝胶支架的基因释放DNA含量于3 d内达高峰,2周内可维持在较高水平。结论携TGF-β3基因自组装肽凝胶基质材料成功合成;此材料可诱导种子细胞定向增殖分化,促进软骨组织修复。
