原发性高血压患者单核细胞TRPC3的表达水平与细胞因子的关系

Abstract
目的:探讨原发性高血压(essential hypertension,EH)患者单核细胞中经典瞬时受体电位通道3(transient receptor potential canonical channel 3,TRPC3)转录水平与单核细胞中白细胞介素1β (interleukin- 1β,IL-1β)和肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)的转录水平以及外周血单核细胞趋化蛋白1(monocyte chemotactic protein 1,MCP-1)浓度的关系.方法:抽取20例原发性高血压患者和20例血压正常的健康体检者的静脉血,分离获得单核细胞,运用实时定量聚合酶链反应法(real-time PCR)分别检测外周血单核细胞中TRPC3、TNF-α和IL-1β的mRNA,用酶联免疫吸附法(enzyme linked immunosorbent assay,ELISA)检测外周血MCP-1浓度.结果:与血压正常对照组比较,原发性高血压组单核细胞中TRPC3、TNF-α和IL-1β的mRNA表达升高,MCP-1浓度增高,具有统计学差异(P<0.05).单核细胞中TRPC3的mRNA水平与TNF-α和IL-1β的mRNA水平以及MCP-1浓度呈现正相关.结论:原发性高血压作为一个慢性炎症过程,单核细胞中TRPC3表达与细胞因子水平相关,TRPC3表达上调可被认为是单核细胞活化的指标之一.
