Abstract
目的探讨肿瘤增殖型腺病毒Ad-delE1b55KD-shRNA/survivin-EGFP介导的针对survivin的高选择性RNA干扰对在体结肠癌细胞HT-29增殖和凋亡的影响。方法用携带针对人survivin的shRNA的肿瘤增殖型腺病毒Ad-delE1b55KDshRNA/survivin-EGFP转染结肠癌细胞HT-29裸鼠移植瘤模型,以携带相同shRNA的复制缺陷型腺病毒和脂质体载体为对照,检测移植瘤体变化,以及TUNEL法检测肿瘤细胞凋亡率。结果 Ad-delE1b55KD-shRNA/survivin-EGFP介导的高选择性RNA干扰使结肠癌裸鼠移植瘤的瘤体减小、移植瘤细胞凋亡率显著增加,且高于目前常用的复制缺陷型腺病毒载体和脂质体载体(P<0.05)。结论 Ad-delE1b55KD-shRNA/survivin-EGFP介导的高选择性RNA干扰能高效地诱导结肠癌细胞凋亡并抑制其增殖。
