Abstract
目的:探讨脐带血干细胞体外诱导分化为心肌样细胞的动作电位、持续性钙电流特性,以对其进行电生理学鉴定.方法:用密度梯度离心和贴壁法筛选脐带血单个核细胞,培养于DMEM低糖培养基,经5-氮杂胞苷诱导为心肌样细胞,在诱导的不同阶段使用膜片钳技术检测其动作电位、持续性钙电流,并以大鼠心室肌作为对照.结果:脐带血干细胞体外诱导分化为心肌样细胞具有心肌特异性动作电位、持续性钙电流,其心肌特性伴随诱导后培养时间延长日趋典型.结论:脐带血干细胞体外诱导分化为心肌样细胞具有心肌细胞的电生理特性.
