流感疫苗脂质体冻干粉的制备及其免疫原性

Abstract
目的:制备流感疫苗脂质体冻干粉,并考察其粒径分布、包封率及免疫原性。方法:以大豆卵磷脂、胆固醇为原料,采用薄膜蒸发法,加入纯化流感病毒裂解疫苗制备脂质体,加入冻干保护剂冻干。在光学显微镜下观察其形态,并在透射电镜下测定其平均粒径和粒径分布,以Lowry法测定其包封率。小鼠肺部给药后血凝抑制法测定抗体滴度以考察其免疫原性。结果:所制备的流感疫苗脂质体冻干粉复溶后呈圆形和椭圆形,粒度分布均匀,平均粒径约为2.14μm,包封率大于80%。肺部免疫后抗体滴度较腹腔免疫高。结论:所制备的流感疫苗脂质体包封率高,粒度分布均匀,小鼠肺部免疫效果好。流感疫苗脂质体肺部免疫是一种很有潜力的免疫途径,有待进一步研究。
