In recent years, we have dedicated our research efforts to addressing the pressing public health issue of age-related degenerative disc diseases, with a particular focus on the field of skeletal regeneration and the challenging clinical treatment aspects. Through the integration of cutting-edge multidisciplinary technologies, we have undertaken a series of innovative research projects in the field of spinal regenerative medicine. The key highlights of our work include:
Seed Cells: We have successfully generated the first human single-cell atlas of intervertebral discs, shedding light on the cellular heterogeneity within this tissue. Notably, we have identified a novel population of PROCR+ intervertebral disc progenitor cells with remarkable regenerative potential. This groundbreaking discovery was featured as a cover article in the esteemed journal Bone Research (2021). Additionally, we have constructed a comprehensive single-cell database of intervertebral disc degeneration in large animal models, unraveling the evolutionary patterns of PROCR+ intervertebral disc progenitor cells throughout the degenerative process (Cell Proliferation, 2023).
Cellular Regulation: We have pioneered the development of advanced technologies for regulating the fate of stem and progenitor cells. By synergistically incorporating mechanical, biological, and materials science factors, we have achieved spatially and temporally regulate cell behavior. This innovative approach has facilitated long-term cell survival and directed differentiation towards the nucleus pulposus lineage, culminating in establishing highly functional intervertebral disc-like organoids (Advanced Healthcare Materials, 2019; Stem Cells International, 2016;2018).
Tissue Regeneration: We have engineered several novel biomimetic hydrogel scaffolds with intricate microstructural features(Biomaterials, 2017; ACS Macro Letters, 2019; ACS Biomater-Sci&Eng, 2019). One scaffold exhibits exceptional mechanical properties and biological activity, meeting the stringent requirements for successful intervertebral disc repair. Encouragingly, preliminary in vivo studies conducted in large animal models have demonstrated promising outcomes in intervertebral disc regeneration (Biomaterials, 2017).
Collectively, these works have laid the foundation for a comprehensive closed-loop technological system for intervertebral disc regeneration. Through preclinical experiments, we have made strides toward achieving the ultimate goal of restoring intervertebral disc function. Our research endeavors provide theoretical insights and practical strategies for the treatment and regeneration of degenerative disc diseases.
