My long-term goal is to establish a successful career as a physician-scientist performing basic and translational research to improve outcomes in head neck and lung cancer, and continue to be actively involved in innovative clinical trials. As an MD/PhD, my initial research training at MD Anderson in the laboratory of Dr. Jean-Pierre Issa introduced me to concepts in cancer epigenetics and targeted therapy. After my medical training, I was extremely fortunate, while performing my oncology fellowship training at MD Anderson, to have the opportunity to work with the group of Dr. Kurie. This allowed me to develop my first independent project, in the area of lung cancer. This project originated in the analysis of highly metastatic versus less metastatic lung cancers emerging in a KRAS/p53 mouse model of lung cancer, and was successfully funded by a YIA fellowship from the ASCO. During my attending faculty position at Fox Chase Cancer Center, I partnered with Dr. Golemis, a highly skilled translational investigator with expertise in the study of cancer signaling and tumor progression, who initially helped develop Musashi-2 project, in work initially funded by the ACS pilot funding. I discovered an important and unexpected role for the Musashi-2 (Msi2) RNA-binding protein (RBP) as an oncogenic translational regulator in lung cancer, which promotes metastasis. This successful project was published in PNAS in 2016, funded by the 2015-2016 DOD Career Development Award, and resulted in a Ph.D. thesis defense by Dr. Deneka.
Complementing my laboratory activities, I have been actively involved in over 20 therapeutic trials for lung cancer patients at Fox Chase, including innovative immunotherapy trials, and trials targeting patients with mutant EGFR and ALK/ROS1. I served as active member of Research Review Committee (RRC) and engaged in translational work as a senior author on a translational genomics paper. I also connected with Synta Pharmaceuticals, allowing me to be the first academic laboratory with access to the promising compound STA-8666, which is a first in class member of new class of HDC conjugate drugs platform. Based on the work with this highly promising compound, high preclinical activity and low toxicity profile in small cell lung cancer models was seen. Anticipated Phase I/II trials with this compound are planned for 2021-2022. In late 2015 and the first half of 2016, because of a family issue, I temporarily transferred to the UNM Comprehensive Cancer Center where I was actively involved in several innovative research trials in lung cancer, served as a Chief of Thoracic Medical Oncology co-leader for the Lung Cancer Working Group and continued active laboratory research.
Since 2016 at Fox Chase and 2020 at Northwestern, I continued exploring exciting topics in lung cancer research, such as MSI2 regulated signaling pathways in collaboration with Dr. Lengner at UPenn and Dr. Golemis, using in vitro models and transgenic Cre-Lox Kras-P53-Msi1/2 mouse model of lung cancer. I have partnered with Dr. Karanicolas in developing novel specific MSI2/1 inhibitors for lung cancer. My lab is currently NIH R01-funded. In addition, in 2019, I received V Foundation award for the correlative science proposal for a clinical trial. Since 2020, my lab has been also focusing on defining the role of NSD1/2 histone methyltransferases in HNSCC and KRAS metastasis drivers in NSCLC. I continue pre-clinical research to advance our understanding of the role of promising candidate oncogenic proteins in cancer biology and signaling, and to probe them as biomarkers.
