Bio
Wayne Tilley is the inaugural Director of the Dame Roma Mitchell Cancer Research Laboratories, Adelaide Medical School, University of Adelaide, Australia (2002- current). His research program is broadly focussed on hormonal carcinogenesis, with an emphasis on mechanisms of sex hormone signalling and the emergence of resistance to hormonal therapies used in the treatment of breast and prostate cancer. In the early 1990s he established his own breast and prostate cancer research program at Flinders University, South Australia. In collaboration with Professor Marshall, he established the Flinders Cancer Centre at Flinders University / Flinders Medical Centre in the mid 1990s. During that time at Flinders University he developed an internationally recognized research program on sex steroid hormone action in breast and prostate cancer, making a major contribution to understanding mechanisms of resistance to endocrine therapies. He is renowned for leveraging commonalities between these two diseases to advance knowledge and new treatment strategies, evidenced by being PI on the first joint NBCF and Movember funding initiative in Australia to transform treatment for endocrine resistant breast and prostate cancers. His research has demonstrated the oncogenic potential of the AR in prostate cancer and, paradoxically, a tumour suppressor role in breast cancer. A current major research focus is the development of novel AR target therapies for both cancers. More recently, his research has highlighted the importance of cross-talk between progesterone, androgen and estrogen receptors in breast cancer to modulate disease outcomes. As a visiting scientist at Cancer Research UK (CRUK) Cambridge Institute, University of Cambridge (2012-2014) with A/Prof Jason Carroll, Prof Tilley developed invaluable new molecular and proteomic technologies to better interrogate steroid receptors in breast and prostate cancer. This research, coupled with a unique ex vivo preclinical model developed in his laboratory to assess the efficacy of new drugs and to identify biomarkers of treatment response (Clin Canc Res 2012; Cancer Discov 2012; Nature 2015), contributed significantly to a successful multidisciplinary international grant from the US Department of Defense aimed to take two new AR targeting drugs into the clinic for endocrine resistant metastatic prostate cancer. He has published over 200 peer-reviewed papers, including his landmark publications on the AR including “Characterisation and expression of a cDNA encoding the human androgen receptor” (PNAS 1989), “Prostate cancer in a transgenic mouse” (1995), “Medroxyprogesterone acetate therapy in advanced breast cancer: the predictive value of androgen receptor expression” (JCO 1995), “Mutations in the androgen receptor gene are associated with progression of human prostate cancer to androgen independence” (Clin Cancer Res 2006) and “Mutation of the androgen receptor causes oncogenic transformation of the prostate” (PNAS 2005) and "The androgen receptor is a tumour suppressor in estrogen receptor positive breast cancer" (Nature Med 2021) continue to be highly cited.
