Susan Guttentag is a phyician-scientist with clinical expertise in neonatology, and research expertise in lung biology, especially alveolar epithelial cell biology and genetic diseases that promote alveolar epithelial cell injury.
Lung surfactant plays an important role in lung disease. Surfactant is a soapy material made by cells in the small airspaces of the lung, the alveolar type 2 cells, to maintain inflation of airspaces during the breathing cycle. Diseases of surfactant fall into 3 categories: developmental, genetic, and acquired. The immature lungs of premature infants are unable to make sufficient amounts of surfactant, and these infants develop Respiratory Distress Syndrome (RDS) as a result of this developmental problem. Genetic defects in components of surfactant, such as Surfactant protein B (SP-B; SFTPB), can result in poor quality surfactant despite normal lung development. In addition, surfactant components can be inactivated due to process like inflammation, oxidation, and edema, and can thereby contribute to lung dysfunction in these acquired lung diseases. Dr. Guttentag's research focuses on the alveolar type 2 cells in the developing and injured lung, and how abnormal development and injury impact the functions of this important cell.
We use powerful in vitro models of alveolar type 2 cells to understand the physiology and pathophysiology of these cells. Prior work focused on the biosynthesis of surfactant components, and more recently the development of lamellar bodies, the surfactant storage organelle in type 2 cells. We have used gene editing in cell models of type 2 cells in addition to RNAi. We also utilize mouse models of genetic diseases known to perturb type 2 cell functions, enabling us to better understand the transcriptional, genetic, and proteomic basis of development of the alveolar epithelium. Through collaborations with Darrel Kotton's laboratory at Boston University, we now utilize human iPSC differentiation into alveolar type 2 cells to provide important translational experimentation.
