My research study was focused to explore adaptation of cluster bean [Cyamopsis tetragonoloba (L.) Taub.] towards drought. We elucidated the role of anatomy and the lignin biosynthesis pathway in drought-tolerant and sensitive varieties of cluster beans in water shortage by using the traits of anatomy, biochemistry and gene expression of genes involved in the lignin biosynthesis pathway.
I have used a system biology approach to analyze water stress in different varieties of guar at the level of physiology, anatomy and molecular biology in my PhD work. I have worked extensively with most of the physiological measurements such as Gas-exchange systems (worked with LiCor-6400 and LiCor-6800, Lincoln, Nebraska, USA) used to measure gas exchange and chlorophyll flourescence, Spectroradiometer (LiCor-1800) to monitor leaf reflectance spectra, Water potential system and Osmometer used to study plant water status; and Chlorophyll fluorescence with Pulse Amplitude Modulator (Imaging PAM and DUAL PAM-100) to analyze the functioning of PSII and PSI along with several biochemical estimations like membrane integrity, plant pigments, proline, total phenolic content (TPC), lignin accumulation. I also handled optical microscope (DM2500), Microtome and thermocycler. Apart from this, I have also studied and handled various plant systems like Arabidopsis, Cotton, Rice, Wheat, Tomato, Chickpea, Soybean and Leucaena leucocephala.
