Bio
I was awarded a BSc with a double major in Molecular Biology and Biomedical Science with 1st Class honours by Murdoch University in 2004. I was awarded my PhD by The University of Western Australia in October of 2010. I had my children while completing my PhD, and have had significant career disruptions. Thus, I have ~8 years experience post-PhD.
During my PhD I created a biobank of samples from ~350 patients with Paget's disease of bone. I performed sequencing on various genes of interest including SQSTM1/p62, DC-STAMP, RANK, and RIN3. I later performed whole exome sequencing and analysis on the particularly strong families within our cohort.
I was recruited on a joint appointment by Murdoch University and the Perron Institute for Neurological and Translational science as a Senior Research Fellow and Lab Head to move my Functional genomics group to this Institute in 2020.
I have been awarded ~$5M AUD in funding since my PhD was awarded. Most as the Principle Investigator.
I have extensive functional genomics experience, characterising the effects of disease-associated mutation on cellular and protein function. My laboratory skills include genomic and other DNA extractions, RNA and protein extractions, PCR/RT-PCR/q-PCR, Sanger sequencing, genotyping, genotype/phenotype correlation analysis, cell signalling assays, cell differentiation (precursor cells to various bone cell types and human induced pluripotent stem cells into neuronal cell types), bone resorption assays, western blots, confocal microscopy imaging and analysis, protein-protein interactions (e.g., ELISA GST/His pulldowns, co-immunoprecipitations, BRET, co-localisation analysis using Imagej/Fiji, proteomics), autophagy and proteostasis/proteinopathy assays, antisense oligonucleotide RNA transcript modulation, plasmid cloning, viral transduction, database curation and data analysis/statistics.
My research focus expanded ~10 years ago to include neurodegenerative diseases. I focus particularly on the causes of dementia including Alzheimer's and frontotemporal lobar degeneration (FTLD/FTD) and chidlhood dementia, motor neuron disease (MND/ALS). However, I have quite broad interests and keep up to date with the literature on spinal muscular atrophy, vascular dementia, dementia with Lewy bodies, Huntington's disease, Parkinson's, Inclusion Body Myositis and Multiple Sclerosis. I have been a guest lecturer in neurodegeneration at the University of Western Australia since 2015.
I am an expert on the roles of mutant/misfolded/aggregated, or over-expressed, SQSTM1/p62, TDP-43, SOD1, TBK1, FUS, and C9orf72 in ALS/FTD, and how proteostasis (ubiquitin-proteasome system, autophagy and the heat shock proteins and other chaperones) dysfunction and other autophagy receptors (VCP, UBQLN2, OPTN) contribute to ALS/FTLD.
I contribute to these fields by regularly reviewing journal articles, abstracts, grant and fellowship applications locally, nationally (Australia) and internationally. I am also a long-standing committee member and secretary for numerous associations.
I am a passionate mental health advocate, and have been the Mental Health First Aider in my centre since 2020. I have detailed knowledge and experience engaging with individuals living with bipolar disorder, borderline personality disorder and drug-induced psychosis., as well as the difficulties and the positives of living with autism and attention deficit disorder/ADHD. I am also very interested in fibromyalgia and chronic pain research.
